A Cysteine Desulfhydrase Specific for D-Cysteine from the Green Alga Chlorella fusca

چکیده
A cysteine desulfhydrase was purified 110-fold from the green alga Chlorella using conven tional techniques. The isolated cysteine desulfhydrase was specific for D-cysteine having no activity towards L-cysteine. Dand L-cysteine desulfhydrase activities can be separated using DEAE-cellulose chromatography techniques. The isoelectric point o f this enzyme was deter mined to be around a pH o f 4.5 using a chromatofocussing column. The pH-optim um for the D-cysteine desulfhydrase was found to be in the range o f 8.5 to 9 and the apparent for D-cysteine was determined to 0.16 m M . The enzyme was active without addition o f metal ions and EDTA or citric acid did not inhibit this activity.

متن کاملFormation of Elemental Sulfur by Chlorella fusca during Growth on l-Cysteine Ethylester.
During growth on l-cysteine ethylester, Chlorella fusca (211-8b) accumulated a substance which contained bound sulfide, which could be liberated by reduction with dithioerythritol (DTE) as inorganic sulfide. This substance was extracted with hot methanol and purified by thin layer chromatography. This substance liberated free sulfide when incubated with mono- and dithiols, and thiocyanate was f...
متن کاملBiodegradation of bisphenol A and disappearance of its estrogenic activity by the green alga Chlorella fusca var. vacuolata.
Bisphenol A (BPA) is known as an endocrine disruptor and often is found in landfill leachates. Removal of BPA by green alga, Chlorella fusca, was characterized, because we previously found that various phenols were well removed by this strain, including BPA. Chlorella fusca was able to remove almost all BPA in the concentration range from 10 to 80 microM for 168 h under continuous illumination ...

In the unicellular green alga Chlorella sorokiniana (211/8 k), the protein O-acetylserine(thiol)lyase (OASTL), representing the key-enzyme in the biosynthetic cysteine pathway, was isolated and purified to apparent homogeneity. The purification was carried out in cells grown in the presence of all nutrients or in sulphate (S) deprived cells. After 24 h of S-starvation, a 17-fold increase in the...
